Earlier this year, Tonum Health put its natural weight loss supplement, Motus, to the test in human clinical trials.
The goal: see how Motus stacked up against the leading GLP-1 medications in both total weight loss and fat loss vs. lean mass retention.
The results? Motus tied semaglutide at six months for total body weight loss, but significantly outperformed GLP-1s when it came to preserving lean muscle and targeting fat loss.
From a Personal Quest to a Scientific Breakthrough
Since 2016, Tonum has been conducting research at Duke University delivering compelling results in cognitive health and metabolic function.
The journey began as a personal mission: Tonum’s founder sought to protect his son from early-onset Alzheimer’s using natural compounds. Unexpectedly, the research revealed strong weight loss effects during preclinical studies. The Motus Human Clinical Trial
In early 2025, after nine years of animal research, Tonum launched its first 100-patient human trial. Participants received Motus, along with access to the MyNetDiary app for food and exercise along with a smart body-fat scale. For the first three months, participants were encouraged to log meals daily and weigh in monthly.
3 Months: Participants lost an average of 5.7% of total body weight, slightly better than semaglutide benchmarks. Footnote - Interventions delivering >5% is considered a clinical success for prescribed medications. But Tonum did not sit on its success and extended the study to 6 months.
6 Months: Roughly half of the participants opted to continue for another three months. By month six, average weight loss reached 10.4% of body weight and 90% of that came from fat, not lean mass. The outcome tied with semaglutide in total weight loss and directly addressed one of the most widely recognized drawbacks of GLP-1s: loss of muscle mass.

Not all participants in the six-month extension continued daily MyNetDiary app usage, but those who did showed modestly better results. More importantly, participants consistently reported that food logging helped them earlier in the study understand eating patterns and make better choices.
